I do that with several Yahoo groups that I moderate, and it keeps spam to almost nothing. Spammers rarely bother to post nicely once to get off moderation.

Hi guys!
First post here... I'm using Math Comment Spam Protection. Basically asks the visitor to solve a simple math question before the comment is posted. Simply to use, just activate it, go to Theme Editor, open up the Comments.php, and paste a single line of code to where you want to put it. I placed mine after the box requesting for the author's website. It's quite commonly used, and rather effective too. Spam went down from 200 per month to only around 40. Those that did to get through seems to all come from the same source, but Askimet got them anyway. 2 layer protection, works well for me!
